Sanitary selection is one of the two elements of clonal selection and is carried out in parallel to the agronomic selection.
N.B.: Only the sanitary selection phase is carried out for the rootstock varieties.

Detection methods
  1 - BIOLOGICAL TESTING : INDEXING



This involves grafting a scion of a clone to be tested onto a cutting of an indicator variety which will present the symptoms of the virus in question if it is present.



Indexing can be done with wood grafting (3 years) or greengrafting (6 to 12 months). This is the method of reference for clonal certification.

2 - SEROLOGICAL TEST - ELISA
(Enzyme Linked Immuno Sorbent Assay)

Immuno enzymatic technique based on an antibody (serum), antigen (virus) reaction.

The ELISA test is used as a control test for monitoring the sanitary status during the propagation process.

3 - MOLECULAR TEST - PCR

Virus detection using specific molecular markers (primers) by genomic amplification after reverse transcription (RT –PCR) or immuno-capture reverse transcription (IC-RT-PCR).


  Treatment Method


It is mainly used for varieties from which healthy clones cannot be obtained by sanitary selection or for clones which carry viruses but which present a special agronomic interest.

Removal of the apex from a virus carrying plant and grafting in vitro onto a hypocotyl of a seedling plant.
